Analysis of RAD54L Gene (Mutation) by RT-qPCR (CAT#: STEM-MT-0106-LGZ)

Introduction

Also known as: HR54; hHR54; RAD54A; Hrad54<br />The protein encoded by this gene belongs to the death-like helicase superfamily, which is similar to Saccharomyces cerevisiae Rad54, which is known to be involved in DNA homologous recombination and repair. This protein has been shown to play a role in homologous recombination-associated repair of DNA double-strand breaks. Binding of this protein to double-stranded DNA causes DNA topology changes, which are thought to promote homologous DNA pairing, and stimulate DNA recombination. Alternative splicing results in multiple transcript variants encoding the same protein.




Principle

Quantitative reverse transcription PCR (RT-qPCR) is an experimental method applied to PCR experiments using RNA as the starting material. In this method, total or messenger RNA (mRNA) is first transcribed into complementary DNA (cDNA) by reverse transcriptase. Subsequently, qPCR reaction was performed using cDNA as template.

Procedure

1. Sample processing and preparation of PCR reaction system.
2. Add the amplification template, cover the PCR reaction cover, mix well, centrifuge at low speed instantaneously, and transfer to the PCR instrument.
3. Set the program for PCR amplification.
4. Data analysis.
