Gene targeting is the targeted integration of exogenous genes into a certain site on the target cell genome through homologous recombination technology, so as to achieve the purpose of targeted modification and transformation of a certain gene on the chromosome. Gene targeting technology has been widely regarded as an ideal method for specifically modifying and transforming the genetic material of organisms.
ES targeting is to perform DNA homologous recombination in mouse embryonic stem cells (ES cells), reinject ES cells into the blastocoel to form chimeric embryos, and develop chimeric mice in pseudopregnant mice. The chimeric mice are then mated with wild-type mice, thereby passing on the genetic information in the ES cells to the offspring. Its advantage is that it is precise and free of off-targets, and can carry out various complex genetic modifications, which is the gold standard in the industry.
