Description

PROTEINDEX™ Dextrin Sepharose 6FF is a chromatographic medium for the purification of proteins fused to maltose-binding protein (mbp-tagged protein). Recombinant proteins with the MBP tag usually have higher expression levels and higher solubility. Proper folding of the attached protein was also shown to be facilitated by the MBP tag. Since MBP increases solubility, this tag is useful for recombinant proteins that tend to accumulate in an insoluble form (inclusion bodies).

Dextrin is the polymer product of amylose after amylase hydrolysis. Compared with amylose, which is easily degraded by amylase, dextrin is very stable. When dextrin was immobilized on the resin as a ligand substrate to purify the MBP tag, it had consistently strong affinity uptake even after repeated use.

Affinity purification using fast flow of Dextrin Sepharose 6 under physiological conditions and gentle elution using maltose preserves the activity of the target protein. These mild conditions can even purify intact protein complexes.

The base matrix of Dextrin Sepharose 6 Fast Flow is a firm, highly cross-linked 6% agarose. The cross-linking of bases has been optimized, which has high physical and chemical stability and high specific binding.

Our proprietary ProteIndex™ conjugation technology ensures stable immobilization of active ligands and minimal ligand leakage under a wide range of purification buffer conditions. This resin can be reused multiple times to purify the same target protein.
