Cluster of Differentiation 155 (CD155), also known as poliovirus receptor (PVR), is a type I transmembrane glycoprotein belonging to the immunoglobulin superfamily. CD155 is commonly referred to as the poliovirus receptor due to its involvement in the cellular entry of the poliovirus in primates. Its normal cellular function is to establish intercellular adhesive connections between epithelial cells. CD155/PVR was initially isolated based on its ability to mediate the attachment of the poliovirus to host cells. The full-length (or CD155α isoform) is synthesized as a 417-amino acid (aa) precursor, which includes a 20 aa signal sequence, 323 aa extracellular region, 24 aa transmembrane segment, and a 50 aa cytoplasmic tail. The extracellular region comprises an N-terminal V-type and two C2-type Ig-like domains. CD155 is a transmembrane protein with three extracellular immunoglobulin-like domains, namely D1-D3, where D1 is recognized by the virus. Low-resolution structures of CD155 in complex with poliovirus have been observed by electron microscopy, while high-resolution structures of the D1 and D2 domains of CD155 have been obtained using X-ray crystallography.
