C-reactive protein (CRP) is a member of the pentraxin family of proteins known for their characteristic cyclic pentameric structure. The human CRP gene encodes a 224-amino acid precursor. The mature human CRP protein consists of 206 amino acids non-covalently linked to form the pentameric structure. Human CRP shares 71% and 64% amino acid sequence homology with the mouse and rat, respectively. CRP, primarily synthesized by hepatocytes, is a major acute-phase serum protein in humans. IL6, IL1, and glucocorticoids are the major inducers of the CRP gene. The physiological role of CRP is to bind to phosphocholine expressed on the surface of dead or dying cells (and certain types of bacteria) to activate the complement system. CRP binds to phosphocholine on microbes and damaged cells, enhancing phagocytosis by macrophages. As a result, CRP participates in the clearance of necrotic and apoptotic cells. CRP can rise up to 50,000-fold in acute inflammation, such as during infections. It typically increases above normal levels within 6 hours and peaks at around 48 hours. CRP has a constant half-life, so its level is primarily determined by the rate of production. High levels of CRP in humans have been associated with an increased risk of cardiovascular diseases.
