Human Fetuin A, also known as alpha 2-Heremans-Schmid glycoprotein, is encoded by the AHSG gene. It is a major plasma protein and a member of the cystatin superfamily of protease inhibitors. It is expressed by hepatocytes, the principal cell source, and by monocytes/macrophages. The major form of plasma Fetuin A corresponds to a disulfide bond-linked two chains derived from the single chain. It is highly expressed in embryonic cells and adult hepatocytes, and is expressed to a lesser extent in monocytes/macrophages.
Human Fetuin A has several functions. It is a negative acute-phase protein with normal circulating levels in adults (300-600 μg/mL), which decrease significantly (30-50%) during injury and infection. It enhances the entry of cationic inhibitors into macrophages and inhibits both insulin receptor autophosphorylation and unwanted calcification. Additionally, it can act as a natural antagonist against specific TGF-β and BMP signaling proteins.
Recombinant Human Fetuin A is a heterodimeric protein containing a 282 amino acid A chain and a 27 amino acid B chain. The calculated molecular weight of Recombinant Human Fetuin A is 32.9 kDa. Due to glycosylation, recombinant Fetuin A migrates at an apparent molecular weight of 45–55 kDa by SDS-PAGE run under non-reducing conditions.
