Description

The Recombinant Human FOLR1 Protein manufactured by Seafrom is suitable for folate binding and cellular uptake.
Folate Receptor 1 (FOLR1), also known as Folate Receptor alpha, Folate-Binding Protein (FBP), or Folate Receptor, is a member of the folate receptor family. Members of this gene family exhibit high affinity for folate and several reduced folate derivatives, facilitating the intracellular delivery of 5-methyl tetrahydrofolate. Mature FOLR1 is a glycosylphosphatidylinositol (GPI)-anchored protein located on the cell surface. FOLR1 is primarily expressed in epithelial cells and significantly upregulated in many cancer cells. FOLR1 is internalized into the endoplasmic reticulum, where it dissociates from its ligand and recycles back to the cell surface. A soluble form of FOLR1 can be released from the cell surface into the serum and breast milk through proteolytic cleavage. FOLR1 deficiency is the cause of Neurodegenerative Childhood Folate Transport Deficiency (NCFTD), a neurodegenerative disorder resulting from early brain-specific folate deficiency.
The lyophilized product can be stored at 4°C for 24 months, while the reconstituted liquid can be stored at -20°C for 6-12 months, avoiding repeated freeze-thaw cycles.

Specification

Expression Host: Human HEK293 cells
Source: Human
Synonyms: FOLR-1, FBP, FOLR
Protein Sequence: The DNA sequence encodes human FOLR1 (UniProtKB - P15328), expressed with a His tag at the C-terminus.
Molecular Weight: Recombinant human FOLR1 protein contains 211 amino acids, with a predicted theoretical molecular weight of 26.3 kD.
Purity: ≥95%, confirmed by SDS-PAGE gel and high-performance liquid chromatography (HPLC) analysis.
Endotoxin: <0.1 EU/μg
Biological Activity: The binding ability is determined using a functional enzyme-linked immunosorbent assay (ELISA) with folate-bovine serum albumin coated at 5 μg/ml (100 μl/well). The optimal binding concentration of recombinant FOLR1 for 50% maximal binding response is approximately 0.8-2nM.
Purification Method: Chromatographic purification
Composition: Sterile PBS (pH 7.4) freeze-dried powder containing 6% mannitol.
