Principle

At 37℃ and pH7.4, ALT catalyzed the amino conversion reaction between alanine and α-ketoglutaric acid to produce pyruvate and glutamic acid. After the reaction time, phenylhydrazine was added to pyruvate to form phenylhydrazone. Phenylhydrazone was reddish brown under alkaline conditions.

Applications

It is used to detect ALT activity in serum (plasma), animal tissues, cells and cell supernatant.

Procedure

1. Prepare standard samples and experimental samples.
2. Add reaction reagents in order for reaction.
3. Measure the absorbance of each tube.
4. Make the mark curve and calculate the result.
