Annexins are a class of calcium-dependent phospholipid-binding proteins. Members of the annexin family contain four internal repeat domains, each of which contains a type II calcium-binding site. Calcium-binding sites are required for annexin aggregation and cooperative binding of anionic phospholipids and extracellular matrix proteins. This gene encodes a different member of the annexin family in which all four homologous type II calcium-binding sites contain amino acid substitutions in the conserved tetrameric core that impair their function. However, structural analysis revealed that the conserved putative ion channel formed by the tetrad core is intact.




Principle

Quantitative reverse transcription PCR (RT-qPCR) is an experimental method applied to PCR experiments using RNA as the starting material. In this method, total or messenger RNA (mRNA) is first transcribed into complementary DNA (cDNA) by reverse transcriptase. Subsequently, qPCR reaction was performed using cDNA as template.

Procedure

1. Sample processing and preparation of PCR reaction system.
2. Add the amplification template, cover the PCR reaction cover, mix well, centrifuge at low speed instantaneously, and transfer to the PCR instrument.
3. Set the program for PCR amplification.
4. Data analysis.
