Analysis of Ca2+-ATPase Activity by Ultraviolet-visible Spectrophotometer (CAT#: STEM-MB-1367-LGZ)

Introduction

Sensitivity: 0.8 U/g
Detection range: 0.8-41 U/g
Precision: inter-lot difference of 8.800000000000001%, intra-lot difference of 4.1%
Detection equipment: Ultraviolet-visible Spectrophotometer
Detection wavelength: 660 nm

Principle

Atpase can decompose ATP to produce ADP and inorganic phosphorus. The activity level of ATPase can be determined by measuring the amount of inorganic phosphorus. In the control system, the activity of calcium ATPase was inhibited, and the determination system did not inhibit the activity of calcium ATPase. The difference of inorganic phosphorus content between the determination and the control system was the inorganic phosphorus produced by calcium ATPase in the incubation time. Determination of calcium ATPase activity by inorganic phosphorus production.

Applications

It is used to detect Ca2+ -ATPase activity in various animal tissue samples.

Procedure

1. Prepare standard samples and experimental samples.
2. Add reaction reagents in order for reaction.
3. Measure the absorbance of each tube.
4. Make the mark curve and calculate the result.

Materials

• Sample Type: various animal tissue samples
