Direct bilirubin reacts with azo reagent under acidic condition to produce azo bilirubin, which has maximum absorption at 565 nm. The content of direct bilirubin in serum can be determined by measuring the absorbance change.

1. Prepare standard samples and experimental samples.
2. Add reaction reagents in order for reaction.
3. Measure the absorbance of each tube.
4. Make the mark curve and calculate the result.
