Principle

5' -NT hydrolyzes hypoxanthine -5 '-monophosphate (5'-IMP) to produce hypoxanthine, which is converted to hypoxanthine in the presence of purine nucleoside phosphorylase (PNP), which is converted to uric acid and H2O2 by the action of xanthine oxidase. H2O2 reacted with chromogen and 4-amino-antipyrine (4-APP) in the presence of peroxidase (POD) to produce colored quinone. The absorbance growth rate was detected at wavelength 550 nm, and the activity of 5 '-nucleotide enzyme was calculated.

Applications

It is suitable for the detection of 5 '-nucleotide enzyme activity in animal tissue samples or serum and plasma samples.

Procedure

1. Prepare standard samples and experimental samples.
2. Add reaction reagents in order for reaction.
3. Measure the absorbance of each tube.
4. Make the mark curve and calculate the result.
