Analysis of Urea (BUN) by Ultraviolet-visible Spectrophotometer (CAT#: STEM-MB-1345-LGZ)

Introduction

Sensitivity: 0.12 mmol/L
Detection range: 0.12-15 mmol/L
Precision: inter-lot difference of 9.9%, intra-lot difference of 4.9%
Detection equipment: Ultraviolet-visible Spectrophotometer
Detection wavelength: 520 nm

Principle

Urea and diacetyl heated condensation in the acidic reaction environment into pigmentogen diazine compound, the depth of color is proportional to the content of urea, because of the instability of diacetyl, so usually in the reaction system by diacetyl monoxime and strong acid action, diacetyl, and then reaction with urea, condensation into red diazine compound.

Applications

Suitable for the detection of urea content in serum (plasma), saliva and urine.

Procedure

1. Prepare standard samples and experimental samples.
2. Add reaction reagents in order for reaction.
3. Measure the absorbance of each tube.
4. Make the mark curve and calculate the result.

Materials

• Sample Type: serum (plasma), saliva and urine
