Analysis of Phenylalanine Ammonia Lyase (PAL) by Ultraviolet-visible Spectrophotometer (CAT#: STEM-MB-1365-LGZ)

Introduction

Sensitivity: 0.78 U/g
Detection range: 0.78-156 U/g
Precision: inter-lot difference of 4.6%, intra-lot difference of 3.1%
Detection equipment: Ultraviolet-visible Spectrophotometer
Detection wavelength: 290 nm

Principle

PAL catalyzes L-phenylalanine to produce trans-cinnamic acid and ammonia. Trans-cinnamic acid has a maximum absorption at 290 nm, and the activity of PAL is calculated by measuring the increase of OD value at 290 nm.

Applications

For the detection of PAL viability in plant tissue samples.

Procedure

1. Prepare standard samples and experimental samples.
2. Add reaction reagents in order for reaction.
3. Measure the absorbance of each tube.
4. Make the mark curve and calculate the result.

Materials

• Sample Type: plant tissue samples
