Principle

Superoxide anion radical is produced by xanthine and xanthine oxidase reaction system. WST-1 (a water-soluble tetrazole salt) can react with the produced superoxide anion to produce water-soluble C19H14IN5O2 dye. When the tested sample contains superoxide anion radical inhibitor, the formation of C19H14IN5O2 dye can be inhibited. When the measured sample contains superoxide anion free radical substances, promote the formation of C19H14IN5O2 dye, through the colorimetric analysis of WST-1 product can calculate the sample inhibition and production of superoxide anion activity unit.

Applications

It is suitable for detecting the activity of superoxide anion free radical in serum (plasma), urine, cells and cell supernatant, various animal and plant tissues and other samples or white blood cells, and drug samples.

Procedure

1. Prepare standard samples and experimental samples.
2. Add reaction reagents in order for reaction.
3. Measure the absorbance of each tube.
4. Make the mark curve and calculate the result.
