Principle

Using thio-oxidizing coenzyme I(S-NAD+) as hydrogen receptor, 3α-hydroxy-steroid dehydrogenase catalyzes the dehydrogenation of bile acid to produce 3-ketosteroid, which converts S-NAD+ into thio-reducing coenzyme I(S-NADH). Using NADH as hydrogen donor, 3α-hydroxy-steroid dehydrogenase catalyzes the production of bile acid from 3-ketosteroid. S-NADH is continuously generated through enzyme cycle reaction. The maximum absorption peak of the substance is 405 nm.

Applications

It is suitable for the determination of total bile acid (TBA) in serum and animal tissue samples.

Procedure

1. Prepare standard samples and experimental samples.
2. Add reaction reagents in order for reaction.
3. Measure the absorbance of each tube.
4. Make the mark curve and calculate the result.
