C-banding (CAT#: STEM-GT-0011-WXH)

Introduction

C-banding is specifically used for identifying heterochromatin by denaturing chromosomes in a saturated alkaline solution followed by Giemsa staining. C-banding highlights centromere-associated heterochromatin and the heterochromatic portion of the human Y chromosome; it is useful in the evaluation of rearranged and marker chromosomes and polymorphic variants of all centromeres, especially those chromosomes 1, 9, and 16.




Principle

Chromosome banding techniques produce a series of consistent landmarks along the length of metaphase chromosomes that allow for both recognition of individual chromosomes within a genome and identification of specific segments of individual chromosomes.
C-Banding technique (also known as CBG-staining) employs mild alkali treatment and Giemsa staining to highlight chromosomal regions known to contain highly repetitive DNA sequences (satellite DNA) such as those located at or adjacent to centromeres of chromosomes (centromeric heterochromatin) and on the distal portion of the Y chromosome. This type of banding is viewed with a bright-field microscope.

Procedure

1. Place air-dried slide of metaphase chromosomes in glass Coplin jar containing HCl.
2. Rinse slide and air dry
3. Immerse one slide at a time in polyethylene Coplin jar containing freshly prepared 5% Ba(OH)2. Incubate 5 to 15 min in a 50°C water bath.
4. Wash slide very thoroughly with water until no white film remains.
5. Incubate in a 60°C water bath.
6. Rinse well in water and air dry.
7. Stain in Giemsa staining solution/Gurr buffer.
8. Rinse well in water and air dry.
9. View and photograph with bright-field microscope.

Microscope, Air-dried slides of metaphase chromosomes, HCl, barium hydroxide, Giemsa staining solution in Gurrs buffer, Glass and polyethylene Coplin jars
