Introduction

Sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE), as one of the polyacrylamide gel electrophoresis techniques, can determine the molecular weight of proteins and separate, analyze, purify, characterize, quantify and prepare small amounts of protein fractions. It has a wide range of applications in protein expression, especially in E. coli expression systems, and has an irreplaceable role.
The principle of SDS-PAGE shows that charged molecules migrate to the electrode with opposite sign when placed in an electric field. The separation of charged molecules depends on the relative mobility of the charged material. Smaller molecules migrate faster due to the lower resistance during electrophoresis. The structure and charge of the protein also affect the mobility. Sodium dodecyl sulfate and polyacrylamide eliminate the effect of protein structure and charge, and proteins are separated according to the length of the polypeptide chain.
