Analysis of Total Superoxide Dismutase (T-SOD) Activity by Enzyme-labeled Instrument (CAT#: STEM-MB-1489-LGZ)

Introduction

Sensitivity: 2.4 U/mL
Detection range: 2.4-61 U/mL
Precision: inter-lot difference of 5.6%, intra-lot difference of 5.5%
Detection equipment: Enzyme-labeled Instrument
Detection wavelength: 540-560 nm

Principle

Superoxide anion radical (O2.-) is produced through the xanthine and xanthine oxidase reaction system, which oxidizes hydroxylamine to form nitrite and produces purplish red compounds under the action of chromogenic agents. When SOD was added into the reaction system, the disproportionation of superoxide anion radical was catalyzed, and the formation of nitrite was reduced, and the purplish red produced became lighter. SOD activity was calculated according to the absorbance difference between the sample tube and the charge.

Applications

It is suitable for detection of T-SOD activity in serum (plasma), urine, cells and cell supernatant, various animal and plant tissues.

Procedure

1. Prepare standard samples and experimental samples.
2. Add reaction reagents in order for reaction.
3. Measure the absorbance of each tube.
4. Make the mark curve and calculate the result.

Materials

• Sample Type: serum (plasma), urine, cells and cell supernatant, various animal and plant tissues
