Analysis of Acid Phosphatase (ACP) by Ultraviolet-visible Spectrophotometer (CAT#: STEM-MB-1335-LGZ)

Introduction

Sensitivity: 0.27 U/100 mL
Detection range: 0.27-40 U/100 mL
Precision: inter-lot difference of 7.1%, intra-lot difference of 2.8%
Detection equipment: Ultraviolet-visible Spectrophotometer
Detection wavelength: 520 nm

Principle

Acid phosphatase can decompose phenylene disodium phosphate under acidic conditions to produce free phenol and phosphoric acid. Phenol reacts with 4-amino-antipyrine in alkaline solution and oxidizes to red quinone derivatives by potassium ferricyanide. The activity of enzyme can be measured according to the shade of red.

Applications

Used to detect the activity of acid phosphatase in animal serum (plasma), body fluid, tissue and cells.

Procedure

1. Prepare standard samples and experimental samples.
2. Add reaction reagents in order for reaction.
3. Measure the absorbance of each tube.
4. Make the mark curve and calculate the result.

Materials

• Sample Type: animal serum (plasma), body fluid, tissue and cells
