Principle

Gsh-px can promote the reaction of hydrogen peroxide (H2O2) and reduced glutathione (GSH) to produce H2O and oxidized glutathione (GSSG). The enzyme activity was expressed by measuring the oxidation rate of the substrate GSH in unit time, and the yellow thio-nitrobenzoic acid anion (TNB) generated by the reaction of the remaining GSH with dithio-di-dinitrobenzoic acid (DTNB). The concentration of the anion was measured to calculate the reduction of GSH. Since GSH can be oxidized in the absence of enzymes (non-enzymatic reactions), the reduction in GSH due to non-enzymatic reactions must be deducted in the final calculation of enzyme activity.

Applications

It was used to detect the GSH-Px activity in serum, plasma, cultured cells, culture medium and animal and plant tissue samples.

Procedure

1. Prepare standard samples and experimental samples.
2. Add reaction reagents in order for reaction.
3. Measure the absorbance of each tube.
4. Make the mark curve and calculate the result.
