Introduction

DNA affinity purification sequencing (DAP-seq) identifies transcription factor binding sites (TFBS) by expressing transcription factors in vitro. It is not limited by antibodies and species, and has the advantage of high throughput. Since its inception, this technology has been widely used in transcriptional regulation and epigenomics research. The principle is that the protein and DNA expressed in vitro are subjected to affinity purification, and the DNA bound to the protein is eluted for high-throughput sequencing.
