Restriction-site Associated DNA sequencing (RAD-seq) (CAT#: STEM-MB-0053-WXH)

Introduction

Restriction-site Associated DNA sequencing (RAD-seq) represents an alternative to whole genome NGS sequencing for the detection, without a preconceived notion, of SNPs and indel variations in DNA. This method consists in the targeted sequencing of regions adjacent to common restriction sites distributed throughout the genome. The use of several restriction enzymes will increase the coverage of the genome, without the need for a reference genome.

Applications

• Identification of genetic variants (SNPs)
• Phylogenetics
• Germplasm assessment
• Population structure
• Linkage and quantitative trait locus mapping
• Genome wide association studies

Procedure

1.Restriction endonuclease digestion
2.P1 Adapter Ligation
3.Sample multiplexing
4.DNA shearing
5.Ampure XP cleanup and size selection
6.End repair
7.3'-dA overhang addition
8.P2 Adapter ligation
9.RAD tag amplification(PCR)
