Principle

β-hydroxybutyric acid was oxidized by β-hydroxybutyric acid dehydrogenase, and NAD+ was reduced to generate NADH. NADH made WST-8 orange color under the action of hydrogen-transmitting substances. The content of β-hydroxybutyric acid was determined by the absorbance value measured at 450 nm, and the absorbance value was positively correlated with the content of β-hydroxybutyric acid.

Applications

It is used to detect the content of beta-hydroxybutyric acid in serum, plasma, urine and animal tissue samples.

Procedure

1. Prepare standard samples and experimental samples.
2. Add reaction reagents in order for reaction.
3. Measure the absorbance of each tube.
4. Make the mark curve and calculate the result.
