Analysis of Mitochondrial Complex IV (Cytochrome C Oxidase) Activity by Enzyme-labeled Instrument (CAT#: STEM-MB-1471-LGZ)

Introduction

Sensitivity: 3.0 U/L
Detection range: 3.0–88.6 U/L
Precision: inter-lot difference of 9.5%, intra-lot difference of 5%
Detection equipment: Enzyme-labeled Instrument
Detection wavelength: 540-560 nm

Principle

Mitochondrial complex Ⅳ can catalyze the oxidation of reduced cytochrome c to produce oxidized cytochrome c, which has an absorption wavelength at 550 nm. Enzyme activity was calculated by measuring the rate of absorbance decline at wavelength 550 nm.

Applications

To detect the enzyme activity of mitochondrial complex Ⅳ(cytochrome c oxidase) in animal tissues.

Procedure

1. Prepare standard samples and experimental samples.
2. Add reaction reagents in order for reaction.
3. Measure the absorbance of each tube.
4. Make the mark curve and calculate the result.

Materials

• Sample Type: animal tissues
