Chromatin Isolation by RNA Purification Sequencing (CHIRP-Seq) (CAT#: STEM-MB-0070-WXH)

Introduction

Chromatin Isolation by RNA purification sequencing (CHIRP-Seq) is a high-throughput sequencing method to discover regions of the genome which are bound by a specific RNA (or by a ribonucleoprotein containing the RNA of interest). ChiRP-Seq works via affinity capture of a target complex of lncRNA and chromatin by tiling antisense-oligos This technique will allow scientists to generate a map of genomic binding sites of several hundred bases very accurately due to high sensitivity and low background.

Applications

Identify RNA-chromatin interactions on a genome-wide scale.

Procedure

1. Cell Collection and Lysis
2. Probe Preparation
3. Immunoprecipitation
4. Elution of RNA and protein
5. PCR detection of RNA
6. Protein Silver Staining-Mass Spectrometry
7. DNA library construction and sequencing
